Abstract

This review highlights a significant role for sphingosine 1-phosphate (S1P) and lysophosphatidic acid (LPA) in many different types of fibrosis and their therapeutic potential. Indeed, the development of effective LPA receptor 1 (LPA1) antagonists and a neutralising anti-S1P antibody that is currently in Phase 1 clinical trials for treatment of age related macular degeneration indicate that translational progress to the clinic is well advanced. However, both LPA and S1P are involved in the multi-process pathogenesis of fibrosis, being implicated in promoting differentiation of fibroblasts to myofibroblasts, epithelial-mesenchymal transition and fibrocyte homing to fibrotic lesions. These processes provide new opportunities for drug intervention targeted at S1P and LPA biology.
