Abstract

Interpretation and generation of biotransformation data, i.e. identification of physiologically relevant metabolites, defining metabolic pathways, elucidation of metabolite structures etc., have become increasingly visible and important to health authorities (HA). Since the introduction of the FDA MIST guidance and the ICH metabolite guidance M3, early information on metabolites structures and their systemic exposure is of high relevance(FDA, 2011; ICH, 2011) Metabolite profiling is defined as the chromatographic separation, quantification, structure elucidation of peaks in a given biological sample derived from an in vitro, in vivo or clinical study. Moreover, metabolite profiling is very time intensive activity, particularly for preclinical in vivo or clinical studies, which have defined limitations on radiation burden and exposure levels. A main limitation is the lack of automation in metabolite profiling, which requires substantial lab capacity, personal attention and resources from laboratory scientists. To help address these challenges and improve efficiency, we have innovated, developed and implemented a novel automation technology linking a HPLC/UPLC-MS to robotic plate handling platform (PlateCrane) which is coupled to an automated fraction collector (Gilson).
