Abstract

To compare the inter-laboratory variability and the influence of different image analysis systems on results obtained in the alkaline single cell gel electrophoresis (comet) assay, in in vivo experiments the genotoxicants EMS and MMS were used. Tissue samples of liver and jejunum from the same animals were split between two laboratories and processed independently including slide evaluation by image analysis. The results correlated very well between the laboratories and demonstrated a low inter-laboratory experimental variability. Furthermore it was shown that neither the use of different image analysis systems nor the staining procedure of DNA (propidium iodide vs. SYBR® Gold) had a considerable impact on the results or the sensitivity of the assay. In addition, a relatively high stability of the staining intensity of propidium iodide-stained slides was demonstrated when the stained slides were stored for over 3 months in the refrigerator. In conclusion, following a thoroughly defined protocol and standardized routine procedures the in vivo mammalian comet assay is a robust assay generating comparable results between different laboratories.
