Legonaridin: a new member of linaridin RiPP from a Ghanaian Streptomyces isolate

Abstract

Linaridins are rare linear peptides that belong to ribosomally-synthesized and post-translationally modified peptides (RiPPs) and only two, cypemycin and SBR-1832, in this family have been identified so far. Legonaridin has been discovered as a new member of linaridin through metabolite profiling and chemical isolation from the soil bacterium Streptomyces sp. CT34, an isolate collected from Legon, Ghana. An MSn-guided genome mining approach allowed identification of the precursor peptide of legonaridin and the associated leg gene cluster. Bioinformatics analysis of the gene cluster suggested that the biosynthesis of legonaridin is different from the ones for cypemycin and SBR-1832. The structure of legonaridin was then established by HRMS combined with bioinformatics analysis, tandem MS, 1- and 2-D NMR spectroscopy. Consistent with the bioinformatics analysis, although legonaridin possesses reminiscent post-modifications, notably dehydrated threonines and an N,N-dimethylated N-terminus, of those seen in cypemycin and SBR-1832, legonaridin is structurally different from these two known linaridins in that it is completely linear with an unmodified carboxylic acid in C-terminus.
