Multiparametric Analysis of Screening Data: Growing Beyond the Single Dimension
Zhang, Xian, Parker, Christian and Abraham, Yann (2014) Multiparametric Analysis of Screening Data: Growing Beyond the Single Dimension. Journal of Biomolecular Screening, 19 (5). pp. 628-639.
Abstract
Advances in instrumentation now allow for screening of multiple readouts such as transcript or protein levels, or multiple parameters derived from images, while advances in profiling technologies are once again reminding us of the complex nature of biology and disease. Taming this complexity requires integration of all dimensions that can be measured rather than the usual single dimensional view that is commonly used. Although some of these methods, such as PCA, are common in microarray analysis or as a QC procedure, we are far from making full use of the statistical and mathematical methods that have been developed over the years and are being used in other fields of science. This review describes a number of the assay systems being used to generate such data sets "en masse" and the methods being applied to their analysis. We also discuss some of the challenges of applying methods developed in other fields to biology.
